Prepare medium anaerobically under 80% N2 + 20% CO2 gas mixture. Add ingredients (except bicarbonate, fructose and reducing agents), boil, cool under N2 + CO2 and dispense anaerobically under same gas. Autoclave at 121°C for 15 min. Add fructose (N2), bicarbonate (N2 + CO2), cysteine (N 2) and sulfide (N2) from sterile, anaerobic stock solutions. Final pH is 5.9. | ||
